2M Imidazole Solution in Water is a concentrated stock solution used primarily in Molecular Biology & Biochemistry to elute His-tagged proteins by immobilized Metal Affinity Chromatography. (IMAC)
Ni or Co- Chelate Bead columns are used to isolate, then elute bound His-tagged proteins using a diluted stock solution of Imidazole between 100-500 mM concentration.

The Binding and Wash Buffer (10-40mM) is used to minimize non-specific binding of containments during purification. pH buffering offers greater stability to proteins and is suitable for enzyme assays and protein purification.
This 0.2 micron filtered stock solution is generally stable but should be protected from light and stored between 2°C to 8°C (refrigeration temperatures).
It can be used in accordance with Sodium Phosphate or Tris Buffers (pH 7.4–8.0) for which the 2M solution can be diluted to maintain pH stability. It can also be used in conjunction with0.5M Sodium Chloride solution to reduce non-specific electro-static interactions.
Specialized purification of proteins by SDS page or analysis protein purity can be optimized with Urea or Guanidine HCL denaturing conditions. Protein crystallization using PEG 8000, imidazole solutions serve to buffer along with Sodium Acetate Tri-hydrate used in specific pH ranges.
Low concentration Imidazole buffer also available.
