Description:

DNA and RNA are extracted using buffered redistilled water saturated phenol.  The extraction of RNA is performed using phenol with a lower pH of 4.5.

DNA is extracted at  a higher pH of 6-8.

Water Saturated Phenol is thus supplied as pH 4.5 and pH 6 and with a saturated Tris buffer solution to adjust the pH, supplied separately.

Tris buffered Pheol pH 8 is also manufactured as a ready to use solution and buffered to pH 8.  Buffered water saturated phenol has a limited shelf life unless it is stored frozen or has 8-Hydroxyquinoline added.

Phenol Solution pH 8 is available with 8 -Hydroxyquinoline, anti-oxidant included.

Phenol/Chloroform/Iso-amyl alcohol is also supplied to enable the partitioning of DNA and RNA precipitating proteins from solution. The protein precipitates at the inter phase.  At pH 7 both DNA and RNA partition in the upper aqueous phase.  Below pH 7 DNA partitions with the protein fraction.

Phenol products are supplied in amber glass in order to reduce photo-oxidation.  They are normally stored in a refrigerator at 4°C.

It is suggested that long term storage of Phenol solution pH 8 is carried out at minus 26°C in a freezer.  Caution should be taken when freezing and defrosting to avoid bottle breakage.  Slowly bring the solution to temperature by placing in a fridge to defrost, always do this with the bottle in its sealed bag provided.
